Myeloid Plasticity of Neutrophil Precursors Contributes to Eosinophil Emergency Hematopoiesis

چکیده
Abstract Eosinophils and neutrophils are heterogenous cells that play diverse roles in homeostasis inflammation. Tissue eosinophil origins poorly understood, posing challenges the functional interpretation of heterogeneous phenotypes. New evidence from our lab shows unexpected hematopoietic plasticity Ly6G(+)IL-5Rα(+) band-stage with potential to contribute lineage. Using flow cytometry multi-omics approaches mouse human tissue samples derived healthy allergic conditions, we tested hypothesis neutrophil contributes diversity We found at least two distinct populations a baseline, one which expressed classical markers (S100A8/9) while other both (MPO, ELANE) eosinophil-specific (EPX, PRG2) granule proteins, indicating an intermediate state. In treated IL-5/G-CSF vitro, subset mixed characteristics acquired granular proteins cell surface (Siglec8, CCR3). asthma model, using scRNA-seq, overlap eosinophils remodeling functions. also identified activated playing unique Our findings implicate emergency hematopoiesis driven by as overlooked process driving persistence Supported grants GSK ISS NIH R01

Hemogenic endocardium contributes to transient definitive hematopoiesis
Haematopoietic cells arise from spatiotemporally restricted domains in the developing embryo. Although studies of non-mammalian animal and in vitro embryonic stem cell models suggest a close relationship among cardiac, endocardial and haematopoietic lineages, it remains unknown whether the mammalian heart tube serves as a haemogenic organ akin to the dorsal aorta. Here we examine the haemogenic...
متن کاملContribution of CD54 to human eosinophil and neutrophil superoxide production.
We have reported that CD54 on eosinophils is involved in eosinophil degranulation. However, the role of CD54 in eosinophil and neutrophil superoxide production is still uncertain. We assessed the effect of CD54 on eosinophils and neutrophils in recombinant granulocyte-macrophage colony-stimulating factor (rGM-CSF)- or phorbol myristate acetate (PMA)-induced superoxide production through CD18. A...
متن کاملHEMATOPOIESIS Dominant Negative Mutants Implicate STAT5 in Myeloid Cell Proliferation and Neutrophil Differentiation
STAT5 is a member of the signal transducers and activation of transcription (STAT) family of latent transcription factors activated in a variety of cytokine signaling pathways. We introduced alanine substitution mutations in highly conserved regions of murine STAT5A and studied the mutants for dimerization, DNA binding, transactivation, and dominant negative effects on erythropoietin-induced ST...
متن کاملHEMATOPOIESIS Changes in Chromatin Organization at the Neutrophil Elastase Locus Associated With Myeloid Cell Differentiation
Neutrophil elastase, proteinase-3, and azurocidin are primary components of neutrophil azurophilic granules and are encoded by closely linked genes (gene symbols ELA2, PRTN3, and AZU1, respectively) in a region of approximately 50 kb. These genes are coordinately expressed in a granulocytespecific fashion, but the mechanisms defining this pattern of expression are unknown. To understand the rol...
متن کاملNeuronal nitric oxide synthase contributes to the regulation of hematopoiesis.
Nitric oxide (NO) signaling is important for the regulation of hematopoiesis. However, the role of individual NO synthase (NOS) isoforms is unclear. Our results indicate that the neuronal NOS isoform (nNOS) regulates hematopoiesis in vitro and in vivo. nNOS is expressed in adult bone marrow and fetal liver and is enriched in stromal cells. There is a strong correlation between expression of nNO...
